Psilocybin Truffle Dosage: Evolute Institute Is The First and Only Retreat in Europe to Test and Measure Your Psilocybin Dose<\/h1>\n

In virtually all retreats, psilocybin truffle dosage is normally estimated from how much the fresh truffles weigh. For example, participants get doses of 30 grams of truffles. The problem: the strength of the same truffles can vary by a factor of 2.5 between batches, meaning with the same weight of truffles, you can end up with over twice as much or as little active psilocybin without knowing it. As the only provider in Europe, Evolute measures the actual psilocybin content of the specific truffle batch used at each retreat and sets every participant's dose in milligrams of psilocybin instead.<\/p>\n

This claim is specific to non-clinical providers: to our knowledge, Evolute is the first non-clinical psilocybin retreat provider in Europe to make measured concentration a standard part of how every participant's dose is set. The assay technology was developed at Friedrich Schiller University Jena with German federal and European Social Fund backing, and is validated to within about 10% of laboratory-grade HPLC.<\/p>\n <\/div>\n\n

Psilocybin concentration testing is the practice of measuring the actual psilocybin content of the specific truffle batch used at a retreat, rather than inferring psilocybin concentration from the batch's weight. Instead of being told “you will take X grams of truffles,” participants have their psilocybin dosage calculated and communicated in milligrams. This page explains why that distinction matters, how the measurement works, and what changes for participants.<\/p>\n\n

How Much Psilocybin Is In a Truffle Dose?<\/h2>\n

That depends on the strain and the batch, and the weight of the truffles will not tell you. A 2024 laboratory study using liquid chromatography-tandem mass spectrometry measured five commercially sourced Psilocybe cubensis<\/em> strains and found average combined psilocybin and psilocin content ranging from 0.88% to 1.36% of dry weight (Goff et al., 2024). A 2022 clinical review names this same variability as one of the practical obstacles to dosing natural products consistently (MacCallum, Lo, Pistawka, & Deol, 2022).<\/p>\n\n

A 2026 peer-reviewed field study shows what that variability means in a live retreat. Pérez Rosal and Faber deployed the same class of colorimetric assay Evolute is adopting at a seven-day retreat in Jamaica with eleven participants, measuring the actual material used in two sessions held 48 hours apart (Pérez Rosal & Faber, 2026). The first session used a 1:1 blend of two strains measuring 6.08 mg of psilocybin per gram. The second used a single, different strain measuring 15.7 mg\/g, a 2.6-fold difference in potency between the two materials.<\/p>\n\n

Because that team measured concentration instead of assuming it, they could weigh each participant's dose to a known milligram target: 15.2-21.3 mg in the first session, and a deliberately higher 47.1-70.7 mg in the second, raised on purpose to offset the acute tolerance that builds after a recent psilocybin session. The deliberate control is the point. Had those same two materials been dosed by weight alone, still the standard across the retreat field, a 2.6-fold potency gap would have passed straight through to participants, unseen and unintended.<\/p>\n\n

Weight has been the only practical proxy available outside a laboratory. It is a reasonable stand-in when nothing better exists. It is still a stand-in, resting on an assumed average potency that a specific batch may not have.<\/p>\n\n

How Does Evolute Measure Psilocybin Truffle Dosage?<\/h2>\n

With a quantitative colorimetric assay, a simplified, field-deployable adaptation of the analysis normally performed by HPLC. A small, homogenised sample of dried truffle material is crushed and mixed with an extraction liquid. The extract is added to a detection solution that develops a colour complex whose intensity is proportional to the amount of active compound present. A photometer then reads that intensity and reports a numerical value. The sample is homogenised precisely because psilocybin is not distributed evenly through fungal material, so a single piece is not representative of the batch.<\/p>\n

Rather than confirming only that psilocybin is present, the test quantifies total alkaloid content, psilocybin plus psilocin (its active metabolite), as a single total psilocybin concentration, across a working range of roughly 0.1-2.5%, with results validated to within about 10% of HPLC.<\/p>\n

The assay was developed at the Friedrich Schiller University Jena Institute of Pharmacy, under a project funded by the German federal government and the European Social Fund and led by Dr. Felix Blei. It has been validated against reference standards at university level and calibrated through instrumental analysis.<\/p>\n <\/section>\n\n

How Is Your Dose Calculated at the Retreat?<\/h2>\n

In milligrams of psilocybin, based on the measured concentration of the specific batch being used at your retreat, rather than as an approximate number of grams of truffle. The facilitation team can then adjust the amount of truffle material up or down to reach an intended, individualised dose, instead of assuming a fixed batch potency.<\/p>\n

Nothing else about the programme changes. The same medical and psychiatric screening, reviewed by a licensed psychiatrist rather than a facilitator. The same 1:2 facilitator-to-participant ratio. The same Medical Doctor physically present on-site throughout the ceremony, not on call. How Does Measured Dosing Compare to Clinical Trial Dosing?<\/h2>\n

Clinical psilocybin trials dose participants in milligrams of synthesised psilocybin, a precisely known quantity, identical from session to session. Retreats using natural truffles have generally not matched that precision, because the variability in the product was assumed rather than measured. Direct concentration testing does not make truffles synthetic or standardised the way a clinical capsule is. It replaces an assumption with a measurement, on the actual batch being used at your retreat.<\/p>\n
